Abstract:
The purpose of this study was to determine whether the curcumin inhibit cancer cell proliferation and metastasis and prevent from breast cancer. Cancer is a life threatening disease which involves the growth of abnormal cell and spread to other parts of body. There are many causes of cancer such as smoking, excess body weight, physical inactivity, heavy alcohol consumption and poor nutrition. There are many types of cancer such as lung cancer, colorectal cancer, prostate cancer and breast cancer. Breast cancer is one of the main cause of death among women as it is considered a wide spread disease.
Different treatment is available in market for breast cancer from medicine to surgical removal of tumor to chemotherapy and radiotherapy. Curcumin is a natural polyphenol and dietary pigment which have both anti-inflammatory and antioxidant properties. It is a bright yellow chemical which is belong to ginger family, Zingiberaceae. It is the active ingredient derived from turmeric (Curcuma longa), which is used to stop the growth of tumor cell. It has antitumoral activities against some antagonistic recurring cancers. It also inhibits chemically induced carcinogenesis in the skin, fore stomach and colon when it is administer during beginning stages. Curcumin is instable and insoluble in water. We also studied the modulating result of curcumin on apoptosis in the tumors. It is used as an herbal supplements, food flavoring, food coloring and cosmetics. There are different pharmacological effects of curcumin such as anticancer, antioxidant, antimicrobial, anti-inflammatory and free radical clearance. It may show highly cytotoxic to some cancer cell lines.
Introduction:
It has been observed
globally that breast cancer is more familiar among all cancers in women. The 2nd
prominent reason of mortality amongst females in the world is breast cancer (Yang
J, Cao Y, Sun J, et al. 2010). Approximately 1.3 million ladies are suffering
from breast cancer and less than 450000 breast cancer related deaths are record
each year. (Yuan X, Chen Q, et al. 2014). It is a heterogeneous disease that
has different biological properties and clinical behavior.
A polyphenol Curcumin
having low molecular weight & it is elicited from curcuma longa L, that is
go to the Zingiberaceae family (Khan ZA, Chiu J, et al 2009). Commonly,
curcumin is regard as more efficient component in turmeric & approximately 2-8%
quantity is used in turmeric preparations. In accordance of number of
researches, there are different pharmacological properties in curcumin such as
anticancer, antioxidant, anti-inflammatory, antimicrobial and free radical
clearance. (Chen Q, et al 2014; Tam K W, et al 2010)
The anti-proliferative
property of curcumin might be recognized to its capacity to control the cell
cycle, & transcription factors, protein kinases together with NF-KB. Curcumin
exhibit a persuasive anti-proliferation result in a melanoma cell line by preventing
the binding activity of NF-KB. Curcumin applied same special effects on AP-1
and EGR (D.R Siwak, K.M Rocco, et al 2007; A.C Johnson, et al 2006).
Curcumin is insoluble and
are not stable in water. Curcumin usage remains limited in anticancer therapy due
to less absorption of this compound. The best useful solvent for curcumin is
DMSO, called cell toxic solution. numerous scientists have attempt to produce
compounds that are related to curcumin, however with greater disintegration
rates in H2O or other appropriate solutions or to produce medicines
that extend the preservation and maintenance of curcumin in the bio environment.
Different preclinical
study focused on the anticancer efficiency of curcumin has been experienced in certain
cancer prototypes with breast cancer. (K. Graham, G. Yan, et al 2013)
Outcome from these
reviews report that curcumin alone, conjugated with targeted and selective compounds
or antibiotics, & is capable to slow down human cancer cell proliferation
and tumor genesis at different molecular levels. (P. Anand, et al 2008; G.
Palma, et al 2013)
Review of literature:
An Impression Of Curcumin
On The Oncogene Proteins Involved In Proliferation And Apoptosis
Curcumin protects the
carcinogens in rodents by slowing down the production of different tumor
causing cells in the culture. In xenotransplant or orthotransplant animal prototypes,
the development of human malignant tumor was repressed via curcumin by itself
or combined with an antibiotic or radiation. (Kunnamakkara AB, Anand P, et al
2008)
Curcumin also involved in G2/M arrest and
death of cells, repressed proliferation of cells by preventing an assembly
dynamics of microtubules. Moreover, cells were accumulate in cell cycle (G1
phase), & curcumin intimidated the zeste homolog 2 gene expression by
stimulating 3 main members of the mitogen activated protein kinase pathway.
(Hua WF, Chen YC, et al 2010)
The apoptosis (death of cells) cause interference in curcumin induced suppression of growing cancerous cell. It has been observed that in cancerous breast cells, curcumin multiplying cell nuclear antigen and p53 mRNAs. Besides this, it was also investigated that curcumin cause inhibition of motility of cancer cells in breast and directly attack by suppressing the role of α6β4 integrin. (Kim HI, Chung J, et al 2008)
Curcumin Impression On Nuclear
Factor-kB
The majority anticancer substitute
trigger on nuclear factor-kB that intervenes in survival, proliferation, and
metastasis of cells. It has been observed that curcumin stop the cancerous
cells migratory activity, proliferative rate, and adhesion.
Human epidermal growth factor (HEGF) 2
is a significant oncoprotein, which is over expressed in around 15-25% of
breast cancers. Moreover, curcumin increased the effectiveness of chemotherapy
for breast cancer. (Husain DM, et al 2011)
Curcumin Effect On Breast Cancer Stem
Cell
Reappearance of cancer and failure in treatment is
basically due to the cancerous stem cells. Even if the majority cancer causing
cells are destroyed by anticancer therapies, the anticancer result of these
treatments will reduce or even vanish after some series. These outcomes show
that cancerous stem cells present anticancer drugs resistance. Consequently,
the recognition & progress of approaches that can successfully assassinate entirely
types of tumor cells, particularly cancer stem cells have developed essential
research issues.
A mouse model trial represented that even if only
curcumin or MMC treatment has been trialed, it showed repressed growth of tumor.
Conjugated treatment with curcumin and MMC reduced size of tumor cell more
extensively than alone treatments with curcumin or MMC.
Furthermore, curcumin might be converse the conflict of BCSCs to MMC. Accordingly, curcumin could recover the therapeutic result of presently existing breast cancer therapies via erupt cancer cells in breast and may possibly give a new and original method for cancer therapy. (Q Zhou, M. ye, et al 2015).
Curcumin Impression On Vascular Endothelial Growth
Factor
Curcumin perhaps give a clinically helpful utensil for
regulator of vascular endothelial growth factor in cancerous cells. It has been
noticed that curcumin inhibits breast tumor angiogenesis. In addition, the epigallocatechin
gallate (EGCG) and curcumin in combined form are useful and efficient in both
in vitro and in vivo prototypes of ER breast cancer. In this methods, the reorganization
of VEGFR-1 possibly will perform a vital function in the antitumor actions. (Scandlyn
MJ, Stuart EC, et al 2008).
Conclusion:
Curcumin has been observed to have beneficial effects on breast cancer. They possess anticancer, anti-inflammatory, antioxidant and free radical clearance properties. It also has antitumorhalavenal activities against some antagonistic recurring cancers. It is able to inhibit chemically induced carcinogenesis in the skin, fore stomach and colon when it is administer during beginning stages. Different studies and experiment proved that curcumin have anticancer effect that helps to prevent from breast cancer. Halaven is a preception medicine used to treat the breast cancer.
